چکیده

DNA replication is a fundamental biological process involving numerous enzymes that help to ensure the maintenance of genomic integrity. DNA polymerase enzymes are key components of this cellular replication machinery. Their primary function is to selectively incorporate correct nucleotide [i.e., a dNTP (where N1⁄4A, T, C or G), 20-deoxyribonucleoside 50-triphosphate] to pair with the template base in accordance with Watson-Crick DNA base pairing rules (i.e., A opposite T or G opposite C). The ability of polymerases to 1

Investigation the Mechanism of Interaction between Inhibitor ALISERTIB with Protein Kinase A and B Using Modeling, Docking and Molecular Dynamics Simulation

The high level of conservation in ATP-binding sites of protein kinases increasingly demandsthe quest to find selective inhibitors with little cross reactivity. Kinase kinases are a recently discovered group of Kinases found to be involved in several mitotic events. These proteins represent attractive targets for cancer therapy with several small molecule inhibitors undergoing different ph...

DNA Polymerase : Multiple Conformational Changes in the Mechanism of Catalysis†

Stopped-flow fluorescence assay was applied to identify conformational changes in the catalytic cycle of DNA polymerase (Pol ), using a synthetic DNA primer/template containing 2-aminopurine (2-AP) at the template position opposite the incoming dNTP. Two phases of fluorescence change were observed in the stopped-flow fluorescence assay of the incorporation of the correct nucleotide dTTP. The ra...

Creative template-dependent synthesis by human polymerase mu.

Among the many proteins used to repair DNA double-strand breaks by nonhomologous end joining (NHEJ) are two related family X DNA polymerases, Pol λ and Pol µ. Which of these two polymerases is preferentially used for filling DNA gaps during NHEJ partly depends on sequence complementarity at the break, with Pol λ and Pol µ repairing complementary and noncomplementary ends, respectively. To bette...

Altered Order of Substrate Binding by DNA Polymerase X from African Swine Fever Virus†

A sequential ordered substrate binding established previously for several DNA polymerases is generally extended to all DNA polymerases, and the characterization of novel polymerases is often based on the assumption that the enzymes should productively bind DNA substrate first, followed by template-directed dNTP binding. The comprehensive kinetic study of DNA polymerase X (Pol X) from African sw...

Understanding the Mechanism Underlie the Antidiabetic Activity of Oleuropein Using Ex-Vivo Approach

Background: Oleuropein, the main constituent of olive fruit and leaves, has been reported to protect against insulin resistance and diabetes. While many experimental investigations have examined the mechanisms by which oleuropein improves insulin resistance and diabetes, much of these investigations have been carried out in either muscle cell lines or in vivo models two scenarios with many draw...
